TELAAH PUSTAKA: PEMBUATAN BIOSURFAKTAN MENGGUNAKAN MINYAK JELATAH OLEH BAKTERI BACILLUS SUBTILIS

Abstract


Limbah minyak jelantah saat ini menjadi isu terbesar di Indonesia, dari hasil survey ditemukan bahwa sebanyak 1.889,506 ton minyak jelantah dibuang di selokan dan tanah setiap minggunya. Dari permasalahan tersebut, biosurfaktan dapat menjadi salah satu solusi untuk meremediasi minyak jelantah. Untuk produksi biosurfaktan dapat mengambil sumber karbon berupa minyak jelantah dengan bantuan bakteri Bacillus subtilis. Tujuan penelitian ini dilakukan adalah untuk mengetahui pengaruh pH dan suhu inkubasi pada produksi biosurfaktan. Penelitian ini dilakukan dengan cara membuatan inoculum atau suspensi Bacillus Subtilis ditempatkan pada media cair NB (Nutrient Broth). Media pertumbuhan terdiri dari Mineral Salt Medium dengan substrat minyak jelatah. Produksi biosurfaktan diamati dengan mengukur tegangan permukaan menggunakan Tensiometer Du-Nouy, aktivitas emulsifikasi, uji enzim lipase dari krt yang didapat, dan produk kasar biosurfaktan. Suhu 37,5°C dan pH 4 dapat menurunkan tegangan permukaan hingga 20 N/m dan mendapatkan indeks emulsifikasi sebesar 69%. Penelitian yang dilakukan dengan suhu 27°C dapat menurunkan tegangan permukaan hingga 25 N/m dan mendapatkan indeks emulsifikasi sebesar 90%. Penelitian yang dilakukan dengan suhu 37°C dapat menurunkan tegangan permukaan hingga 23 N/m dan mendapatkan indeks emulsifikasi sebesar 62%. Dengan dilakukan penelitian ini diharapkan dapat mengurangi limbah minyak jelantah dan didapatkan surfaktan yang bersifat biodregedeble dan ramah lingkungan.
